Abstract

The ability of p53 to promote apoptosis in response to mitogenic oncogenes appears to be critical for its tumor suppressor function. Caspase-9 and its cofactor Apaf-1 were found to be essential downstream components of p53 in Myc-induced apoptosis. Like p53 null cells, mouse embryo fibroblast cells deficient in Apaf-1 and caspase-9, and expressing c- Myc , were resistant to apoptotic stimuli that mimic conditions in developing tumors. Inactivation of Apaf-1 or caspase-9 substituted for p53 loss in promoting the oncogenic transformation of Myc -expressing cells. These results imply a role for Apaf-1 and caspase-9 in controlling tumor development.

  31. Primary MEFs derived from wild-type Apaf-1 −/− Casp9 −/− and p53 −/− mice were prepared as described [M. Serrano et al. Cell 88 593 (1997) and (13)]. Stable expression of c- Myc H- RasV12 and a COOH-terminal GFP fusion of a temperature-sensitive human p53 (Val 138 ) was achieved by retroviral-mediated gene transfer as described [

  33. ] except that MEFs were infected four times at 4-hour intervals. Retroviral vectors were as follows: c-Myc/MarXII-hygro [X. Wang et al. Genes Dev. 12 1764 (1998)] pBabeRas [M. Serrano et al. Cell 88 593 (1997)] and pWZL-GFP p53 V138 (gift of J. Hudson).

For cell viability experiments 2 × 10 5 cells were plated in Dulbecco's modified Eagle's medium (DMEM) supplemented with 10% fetal bovine serum (FBS); 12 hours after plating cells were placed in 0.1% FBS (growth-survival factor withdrawal) 0.2% oxygen (hypoxia) 50 μg of murine TNF-α (mTNF-α) per milliliter (Fig. 1) or in 1% FBS at the restrictive or permissive temperature for p53 expression (Fig. 2). For suspension assays 2 × 10 4 cells per 6-cm plate were resuspended in DMEM containing10% FBS and added on top of solidified 0.5% Noble agar. At various times after each stimulus floating and adherent cells were pooled and analyzed for cell viability by trypan blue exclusion and for apoptosis by 4' 6-diamidino-2-phenylindole (DAPI) staining for condensed chromatin (12).
