Abstract
Immature thymocytes express a pre–T cell receptor (pre-TCR) composed of the TCRβ chain paired with pre-Tα. Signals from this receptor are essential for passage of thymocytes through a key developmental checkpoint in the thymus. These signals were efficiently delivered in vivo by a truncated form of the murine pre-TCR that lacked all of its extracellular immunoglobulin domains. De novo expression of the truncated pre-TCR or an intact αβTCR was sufficient to activate characteristic TCR signaling pathways in a T cell line. These findings support the view that recognition of an extracellular ligand is not required for pre-TCR function.

) / Immunity by Thornell A. P. (1997) - A fragment of DNA encoding the COOH-terminal 51 amino acids of murine TCRβ was obtained by polymerase chain reaction (PCR) from a TCRβ cDNA clone. Oligonucleotides encoding the CD8α leader and the Flag epitope (40) were inserted into an engineered restriction site upstream of the cloned PCR fragment. pTα T which begins nine amino acids on the NH 2 -terminal side of the membrane-proximal cysteine was generated by PCR with reverse-transcribed RNA from the θ8c ( Rag-1 −/− ; p53 −/− ) thymoma. An oligonucleotide encoding the Myc epitope tag (41) was added on the 3′ side of the CD8α leader sequence. Constructs encoding β T and pTα T were sequenced and subcloned into a modified form of the pEF-Boss vector (30) for transfection into the θ8c thymoma (18) and the TCRβ-deficient mutant of Jurkat JRT3.T3.5 (26). For production of the transgenic mice β T and pTα T were cloned into the p1017 expression vector which includes the Lck proximal promoter and the human growth hormone 3′–untranslated region (42).

(1995) - Mice transgenic for β T and pTα T were generated and screened by standard procedures (43) with the use of (C57BL/6 × DBA/2)F2 and (C57BL/6 × CBA/Ca)F2 zygotes respectively. One transgenic line containing about six copies of β T was crossed with a founder containing a high copy number of the pTα T transgene and bred to Rag-1 −/− mice. C57BL/6 mice were used as wild-type controls in all experiments. Rag-1 −/− ES cells (5A137D4) (24) were transfected with expression vectors containing both β T and pTα T in addition to a plasmid encoding resistance to puromycin (PGK-puro). Puromycin-resistant colonies were isolated screened for transgenes by Southern (DNA) blot and microinjected into Rag-1 −/− blastocysts to produce chimeric mice according to standard procedures (43).

- TCRβ-deficient Jurkat cells (20 × 10 6 ) were electroporated in 300 μl of serum-free RPMI 1640 medium containing 5 μg of NFAT-luciferase plasmid (44) 6 μg of v- Ras plasmid and a total of 25 μg of pEF-Boss–based plasmids in a Bio-Rad Gene Pulser set at 240 V and 960 μF. About 6 hours after transfection the cells were counted in a hemocytometer and plated in triplicate into 96-well plates at 200 000 cells per well. At selected times the 96-well plates were frozen at −70°C for later analysis of luciferase activity as previously described (27).

- Single-cell suspensions of thymocytes were stained with the following antibodies (purchased from Caltag Pharmingen Becton-Dickinson or VWR) in phosphate-buffered saline plus 0.3% bovine serum albumin: antibody to CD4 (anti-CD4) (CT-CD4-Tricolor) anti-CD8 [53-6.7–fluorescein isothiocyanate (FITC)] anti-TCRβ [H57-597–phycoerythrin (PE)] anti-CD25 (7D4-FITC) anti-Flag (M2) anti–Myc epitope (9E10) and anti-CD3ε (145-2C11-PE). Jurkat transfectants were stained with Leu 4 (anti-human CD3ε) followed by FITC-conjugated goat antibody to mouse IgG. Stained cells were analyzed with a Becton-Dickinson FACScan and CellQuest software. Dead cells were excluded by gating on the forward and side scatter.
