ATPase and GTPase Activities Isolated from Rat Liver Ribosomes

Abstract

An RNA · protein complex containing ribosomal 5‐S RNA has been isolated by treatment of rat liver 60‐S ribosomal subunits with EDTA. The protein moiety of this complex shows one band in dodecylsulfate gel electrophoresis whose mobility suggests a molecular weight of about 32000. The complex exhibits both GTPase and ATPase activities. The optimal conditions for both enzymic activities have been studied. Kinetic experiments revealed that GTPase and ATPase both had a Km of 3.7 μM, whereas V values for GTPase and ATPase were 5.6 and 2.2 pmol/min, respectively. Inhibition studies have shown that two different enzymatic sites are involved in both reactions.
