The Solubilization of the Cytoplasmic Membrane of Bacillus subtilis by Sodium Dodecyl Sulphate

Abstract

Cytoplasmie membranes of Bacillus subtilis have been solubilized by brief exposure to the anionic detergent, sodium dodecyl sulphate. Complete solubilization was obtained when the relative proportation of detergent to membrane protein was approximately 1:2.The solubilized membrane preparation can be completely freed of detergent by gel filtration. The membrane components do not reaggregate, and membrane phospholipid remains associated with the protein during solubilization and subsequent removal of the detergent.Model experiments showing that enzymes can be selectively solubilized by varying the concentration of sodium dodecyl sulphate, indicate that the solubilization of the membrane preparation by the detergent is not a random disaggregation.
