Abstract

SummaryA detailed procedure is described for obtaining human iron‐transferrin with a purity exceeding 90 % from Cohn Fraction IV‐4. The procedure has been designed for use with convential, large‐scale blood fractionation equipment. Data on yields, purities and composition of fractions from a series of large‐scale runs are given.The preparation of purified iron‐transferrin for intravenous use in humans is described. A study was made of the heat treatment (10 hours at 60° C.) commonly used for inactivating the hepatitis virus. The proposed unit of human transferrin for clinical evaluation contains approximately 5 g. of the purified protein as the iron complex in 50 ml. of a sterile, neutral saline solution.RésuméLes auteurs décrivent dans le détail une méthode pour la préparation de la sidérophiline humaine purifiée à 90% à partir de la fraction IV‐4 de Cohn. La méthode est adaptée aux installations techniques habituelles de fractionnement prévues pour de grandes quantités de sang. Les auteurs fournissent des données concernant le rendement, la pureté et la composition de fraction provenant d'une série de plusieurs lots.Ils décrivent la préparation de la sidérophiline purifiée pour injection intraveineuse; ils étudient le comportement de la préparation après une incubation de 10 heures à 60° pour inactiver le virus de l'hépatite. Pour les besoins cliniques, les auteurs proposent que l'unité de sidérophiline humaine soit constituée par 5 grammes de protéine purifiée contenant le complexe ferrique dilué dans 50 cc de solution physiologique neutre et stérile.ZusammenfassungEs wird ein Verfahren zur Gewinnung von menschlichem Eisen‐Transferrin mit einer Reinheit von über 90 % aus der Cohn'schen Fraktion IV‐4 beschrieben. Das Verfahren wurde den speziellen Bedürfnissen der großtechnischen Plasmafraktionierung angepaßt. Es werden Angaben über Ausbeuten, Reinheitsgrad und Zusammensetzung der Fraktionen einer Reihe von technischen Fabrikationsserien gemacht.Die Herstellung eines gereinigten Eisen‐Transferrin‐Präparates für intravenöse Verabreichung wird beschrieben. Die Frage der Wärmebehandlung dieser Präparate (10 Stunden 60° C) wurde im Hinblick auf die Inaktivierung des Hepatitisvirus untersucht. Die für klinische Zwecke vorgeschlagene Transferrineinheit enthält etwa 5 g des gereinigten Proteins als Eisenkomplex in 50 ml steriler neutraler Salzlösung.
