Abstract
AbstractThe optical rotatory dispersion of a series of proteins was studied and compared by using two improved instrumentations. The measurements were made in the wave length range of 200–400 mμ. The earlier with less perfect instruments obtained results wre corrected. It was found that regarding the rotatory dispersion in far ultraviolet, all native globular proteins can be classified into two groups: (1) those showing a negative COTTON effect with a sharp minimum in the dispersion curves at 233 mμ, and (2) those having in their curves a flat minimum at 220–233 mμ. To the first group belong the proteins which have a strongly negative. MOFFITT‐YANG constant (b0) and whose polypeptide chains probably have the α‐helical conformation to a large extent. The native proteins of the second group (b0 = 0) probably are devoid of the α‐helix or have it only in relatively in significant amount. The amplitude and position of the negative COTTON effect can be applied as an independent criterion for the study of the conformation. The COTTON effect study is especially useful for substances which are available in very small quantities, since the specific rotation measured with the wave length of 233 mμ ranges between −1600 and −12,000°.
