  22. The self‐assembly of the linear constructs is essentially quantitative. Also the self‐assembly of the DNA hexagon is quite efficient. The isolated yield of the hexagon is 60 % whereas the amount of hexagon in the crude mixture is 75 % (evaluated from UV/Vis spectroscopy). Scale‐up of this synthesis is expected to produce more oligomers if the concentration of DNA is increased. However the yield of the DNA hexagon should not be affected if volumes are increased but similar concentrations of DNA are used.

The 1:1 ratio of DNA to gold was verified by 1) UV/Vis quantification of a band solely due to the gold particles versus a band due to DNA; 2) titration experiments of the gold–DNA constructs with their complementary partners which showed that a single hybridized product is only obtained at a 1:1 ratio; 3) silver enhancement staining of the bands under conditions where DNA alone does not stain; and 4) significantly reduced band mobility on native agarose gels compared to DNA alone.
